Will repeatedly playing through the same tomb, killing the same boss, and so on reduce the quality of loot over time? I'd doubt it, considering what I hear about Typhon runs, but I wasn't sure.

Not that I have noticed. I farm some areas over and over for a while (i.e. Nessus - 4 chests there and gorgons, 4 or 5 chests there) to build up some cash reserves and upgrade gear. Though I do think that each chest has a level range that it will drop for.

Oh, cool.
So to chests, the answer is no, and to red bosses the answer is yes?
yup, that sums it up quite nicely.
edit: One minor point though, the loot of red bosses includes the spheres and majistic chests...

That isn't a minor point!
Majestic Chests are like exactly what I was focusing on in this post.
Hmm big point then.... Sorry about the confusion.
By the way the Typhon runs you read about are probably before the 1.20 patch. I believe the loot drop wasn't working as the devs wanted it to, giving the same high loot drop every drop when it was supposed to be that high with the first drop only.
